Research Interests:
The
focus of my research program is the immunopathogenesis and
prevention of human and feline immunodeficiency virus infection
(HIV and FIV, respectively). FIV is a valuable model of human
immunodeficiency virus infection in people and represents
a significant health threat to the feline population world-wide.
We are investigating the role of innate immune defects induced
by HIV/FIV infection. These studies will carefully define
the nature of the innate immune defects with an emphasis on
toll-like receptors (TLR). Modulation of the innate response
through cytokine or TLR manipulation will be addressed as
a strategy to reduce debilitating opportunistic infections
in AIDS patients. These studies will also determine the impact
of a reduced innate immune response on the induction of immunologic
memory. Ongoing vaccine studies seek to employ recombinant
bacteria as vaccine vectors. We are evaluating recombinant
Lactobacillus gasseri and Listeria monocytogenes engineered
to express HIV and FIV structural genes and deliver an Env-expressing
plasmid to antigen presenting cells as a means to provide
effective mucosal and systemic immune responses through oral
immunization.
