Becton-Dickinson FASCan
Has a single argon laser with an excitation wavelength of 488nm. It can provide data for forward scatter (FSC), side scatter (SSC), and 3 colors of fluorescence. The fluorescence wavelengths that can be routinely collected are FL1 (525nm), FL2 (575nm), and FL3 (>650nm). The fluorochromes that are best suited for routine use on the FACScan are:
- FL1: fluorescein, GFP (green fluorescent protein) or some of the fluorescein alternatives such as Alexa 488
- FL2: R-phycoerythrin (R-PE)(Cy3 can also be detected)
- FL3: PerCP or R-PE-Cy5
Becton-Dickinson FACSCalibur
Provides all of the same capabilities as the FACScan. In addition it has a second excitation laser (633nm) and, thus, is capable of analyzing up to four colors of fluorescence. The 4th flourochrome used is usually allophycocyanin (APC).
All analyzers have the capacity to do DNA analysis using Propidium Iodide or other stains excited by a 488 nm laser or 633 nm laser.
Cytek modified BD FACScan
Has been modified with a 635 nm laser and two additional PMTS to provide a total of five colors from the 488 nm and 635 nm lasers. Utilizes FlowJo Collection Software for acquisition and FlowJo for analysis.
Becton Dickinson LSRII
Has three lasers, 488 nm, 635 nm and 405 nm. Can analyze up to nine colors and utilizes BD Diva software for acquisition. Scheduling for the LSRII is available by online invitation calendar.
